    Universal principles of SMC-mediated genome organization
    The Structural Maintenance of Chromosome (SMC) protein family plays a crucial role in chromosome structure and is highly conserved across all life forms. It is involved in various vital biological processes, including chromosome organization, segregation, gene expression, replication, and repair. Yet, the fundamental principles guiding SMC proteins in chromosome formation remain unclear. To explore these principles, we will employ single-molecule DNA tethering imaging to observe real-time interactions between SMC proteins and DNA at a single-molecule level. Additionally, we will study the structural dynamics of SMC proteins using high-speed atomic force microscopy. Our investigation will encompass SMC proteins from different species, aiming to unravel longstanding biological mysteries surrounding chromosome formation.&nbsp;<br /> &nbsp; <hr /><br /> SMC (Structural Maintenance of Chromosome) 단백질군은 염색체 구조를 형성하는 주요 단백질로 &nbsp;원핵생물부터 인간까지 크게 보존되어 있다. SMC 단백질군은 정밀한 염색체 구조 형성 및 딸 세포 분리 작용에 관여함으로 유전자를 세대에 걸쳐서 효과적으로 전달하게 하고, 유전자 발현, 유전자 복제, 유전자 복구 등의 다양하고 중요한 생명현상과 관련되어 있기 때문에, 생물학적으로 중요한 단백질군이다. 하지만, 어떻게 SMC 단백질 군이 염색체 형성을 시키는지에 대한 보편적인 원리는 아직 결론이 나지 않은 상태이다. 본 연구는 SMC 단백질의 보편적인 원리를 연구하고자, 세포 밖에서 단일 분자 DNA 양 끝단 테더링 이미징 기법을 이용해 단일 DNA 위에서 SMC단백질과 DNA 가 어떻게 상호작용하는지를 단일분자 수준으로 실시간으로 관찰한다. 또한, 초고속 액상 원자힘 현미경 (HS AFM)을 이용하여 SMC 단백질의 구조 동역학을 이해함으로 SMC 단백질의 분자 기전을 이해할 것이다. 다양한 종의 서로 다른 SMC를 사용하여 이들 SMC 단백질들의 보편적인 원리를 이해하려고 한다. 이 일을 통해서, 염색체 형성에 관련된 여러가지 생물학적인 오래된 미스터리를 해결하는 데 기여할 것이다.&nbsp;

    Dissecting neural mechanisms of flexible information processing: convergent approaches spanning molecular- and systems neuroscience and computational decoding
    Cognitive flexibility is crucial in daily life as it enables us to flexibly update old knowledge with new relevant information. This higher-order cognition is often impaired in neuronal disorders. Notably, getting away from everyday life, or experiencing novelty, prevents dementia and improves cognitive flexibility. Thus, investigating this natural way of enhancing cognitive flexibility can provide a fresh perspective on treating cognitive deficits, which complements current research on disease models.<br /> Most animal studies, however, rely on simple behavior that does not require flexible information processing. Moreover, current systems neuroscience research is often limited to one brain area or a simple one-projection circuit, lacking holistic approaches to determine complex cognitive processing. Further, although the functional roles of neural activities have been extensively studied, the absence of computational decoding fails to dictate how they process cognition. Finally, the gap between systems neuroscience and decades of molecular research remains unresolved.<br /> I will overcome these hurdles using my interdisciplinary expertise in molecular- and systems neuroscience and computational decoding. With a newly designed behavioral paradigm, my recent corresponding-author publications and unpublished data found that novelty disrupts synchrony across the key learning and memory circuit, including the prefrontal cortex, hippocampus, and ventral tegmental area. Decoding analysis revealed that this effect boosts information updating. This finding seemingly contradicts the common notion that stronger synchrony leads to better performance. Molecularly, I found evidence that activin signaling pathways may mediate these effects of novelty. Here, I propose to outline flexible information processing by integrating my interdisciplinary expertise. This convergent project will challenge the current mechanistic understanding of cognition and establish a new norm in brain research.<br /> &nbsp; <hr /><br /> 사람이 만물의 영장인 이유는 고차원적인 사고가 가능하기 때문이다. 우리는 일상생활에서 얻은 정보를 저장하고, 새로운 정보가 주어졌을 때 이를 수정/보완한다. 이렇게 계속적으로 정보를 저장, 갱신하는 능력, 즉 인지적 유연성은 끊임없이 변화하는 세상에 빠르게 대처하여 생존을 보장하는 기반이 된다. 주목할 점은 인지적 유연성의 감소가 자폐, 조현병 등 여러 정신질환뿐만 아니라, 노화나 수면 부족 상태에서도 나타난다는 것이다. 본 연구는 인지적 유연성의 기전을 분자생물학과 뇌의 학습/기억 회로 내 신호전달 및 이 신호의 수학적 해석을 통합해서 규명하고자 한다. 이 연구를 통해 각 연구 영역별로 단편적으로 이해하던 고차원적 인지능력의 기전을 종합적으로 규명함으로써 인지장애 극복의 토대를 마련하고자 한다.

    Tracing fundamental principle of tissue-specific tumorigenesis
    The enigmatic nature of tissue and cell type specificity of tumor formation across diverse tissues has long posed a formidable challenge in cancer research. This specificity of tumorigenesis suggests that complex intracellular networks, closely intertwined with tissue environments, may govern the predisposition to tumor initiation. However, the remarkable plasticity and temporal fate transitions of epithelial cells present a significant hurdle to study the tumor initiation process using current approaches. Here, we propose an innovative approach, which overcomes the limitations of previous approaches and allows multiplexed tracking of cellular states from virtually every cell in the tissue at any desired time point. By spatially recording cellular states during tumor initiation in time evolution, we will complete the first &lsquo;cellular history map&rsquo; of tumor initiation. This approach will offer invaluable quantitative insights into tumor initiation processes, facilitating the identification of basic principles governing the transformation of cells with oncogenic driver mutations, into actual tumor.&nbsp;<br /> &nbsp; <hr /><br /> 암은 돌연변이의 축적으로 인해 발생한다고 알려져 있다. 하지만 조직에서 암이 되지 않은 상태로 존재하는 암 돌연변이 세포들이 빈번하게 발견되면서, 암 발생과정이 실제로는 각 세포의 상태마다 다를 수 있다는 가능성이 제기되었다. 본 연구에서는 정상 세포가 암세포로 변해가는 과정을 시간과 공간의 측면에서 각각 추적함으로써 암이 발생하는 과정을 정밀하게 들여다보고자 한다. 이를 위해 오가노이드 및 유전공학기술을 활용하여 세포 내에서 일어나는 시계열적 상태 변화와 미세환경을 동시에 기록할 것이다. 이를 통해 암 발생의 근본적인 원인을 규명하고, 새로운 암 예방 전략을 발굴하고자 한다.&nbsp;

    Unveiling the hidden guardians of brain health: Exploring immune cells in non-pathological brain conditions
    In the exciting field of neuroimmunology, there is a remarkable and uncharted area waiting to be explored&mdash;the mysterious role of brain-specific immune cells in maintaining a healthy brain under normal conditions. This discovery presents a paradigm shift, challenging our conventional understanding that immune cells only function fighting against pathogens and tumors. Recent evidence indicates that immune cells possess unique traits in the healthy brain that potentially go beyond their traditional roles in disease contexts. However, there is still much to learn about their functions and communication with brain-resident cells in a healthy brain. In our groundbreaking study, we aim to uncover the functional maps of brain-specific immune cells during normal brain development and explore their unique role in maintaining brain balance. Addressing these key questions, we aspire to redefine the boundaries of neuroimmunology research.<br /> &nbsp; <hr /><br /> 뇌에 존재하는 면역세포가 질병이 없는 건강한 상태에서도 신경세포에 영향을 미칠 수 있을까? 최근 활발히 연구되고 있는 뇌-면역 축 (Brain-Immune Axis)은 주로 질병 상태에서 외부로 부터 유입된 면역세포들이 뇌의 퇴행이나 자가면역 질환에 관여한다는 내용이다. 상대적으로, 건강한 상태의 뇌에 존재하는 면역세포에 대한 기능과 신경세포와의 연결 메커니즘은 아직 잘 알려지지 않았다. 따라서, 본 연구실에서는 혈뇌 장벽 (Blood-Brain Barrier)이 생성되기 전 뇌로 이주한 면역세포들이 신경세포와 같은 뇌 상주 세포들과 어떻게 소통을 하는지 알아보고자 한다. 더 나아가, 이를 통해 뇌에 정착한 면역세포는 질병 상태에서 혈액으로부터 유입되는 면역세포와 달리, 뇌의 정상 신경발생 과정에 영향을 미친다는 가설을 입증하고자 한다.
